Zinc is essential for nail matrix cell division and keratin protein synthesis. It regulates matrix metalloproteinases that remodel the nail plate and supports superoxide dismutase activity that protects nail cells from oxidative damage.
How strong is the research?

Who was studied: Patients whose nail changes came from a diagnosed mineral deficiency.
No human trial with a verifiable DOI testing zinc supplementation for brittle nails as a primary endpoint was found. The best available evidence (Scheinfeld et al. 2007, PMID 17763607) explicitly finds no support for zinc in nail health in well-nourished patients. We cannot stand behind this claim on the evidence we have, so it carries no rating.

Before you take it
- NIH ODS Tolerable Upper Intake Level (UL) for zinc is 40 mg/day for adults aged 19+ (all dietary and supplemental sources combined).
- Antibiotic interaction (asymmetric): zinc chelates tetracyclines (doxycycline, minocycline) and fluoroquinolones, reducing their absorption by 30–50%. Take antibiotics at least 2 hours BEFORE zinc, or 4–6 hours AFTER zinc (NIH ODS).
- Chronic zinc supplementation — particularly above 25 mg/day sustained over months — suppresses intestinal copper absorption via metallothionein induction; monitor serum copper and ceruloplasmin with long-term use.
- Copper deficiency secondary to excess zinc causes microcytic anemia, neurological symptoms, and paradoxically worsens the very hair, nail, and skin complaints zinc is often taken to address.
- Zinc picolinate has very limited dedicated clinical trial data relative to zinc sulfate, gluconate, or acetate; most therapeutic evidence for zinc uses other salt forms, and head-to-head bioavailability advantages of picolinate over other forms remain contested.
- Simultaneous ingestion of iron supplements reduces zinc absorption and vice versa; separate by at least 2 hours when both are prescribed.
- Pregnancy: RDA is 11 mg/day; lactation: 12 mg/day. Supplemental doses approaching or exceeding the 40 mg/day UL are not recommended during pregnancy or lactation.
- PPI and H2 blocker use reduces gastric acid, which impairs ionization and absorption of zinc salts including zinc picolinate; acid-suppressant users may absorb less zinc from supplementation.
